Am 22. November 1963 hält die Welt den Atem an: John F. Kennedy, der jüngste ins Amt gewählte Präsident der USA, wird von mehreren Schüssen niedergestreckt. In Windeseile präsentiert das FBI einen Täter, den Einzelgänger Lee Harvey Oswald. Zwei Tage später wird er von Jack Ruby erschossen. Die mit der Klärung des Falls beauftragte Warren-Kommission kommt zu dem Schluß, daß Oswald der alleinige Täter war. Staatsanwalt Jim Garrison zweifelt die Theorie an und macht sich mit seinem Team daran, das Mordpuzzle selbst zu lösen. Obwohl Zeugen wie die Fliegen unter merkwürdigen Umständen sterben, deckt Garrison ein Komplott auf, an dem CIA, FBI, die Mafia und Präsident Johnson gleichsam beteiligt sind.
